If my wife quit claims her half ownership to me, how does that affect the owner of the other half of the property, her brother? If it does not, must the other half owner agree to or sign the/an agreement or deed? The property is located in Virginia and there is no mortgage or liens on the property.

Your wife's brother need not sing on the deed because he isn't transferring his property share over to you. Only your wife is conveying her part of property. So, such a transfer will not affect the brother.
